No, it doesn't.  It may mean "no GTK+", but GLib doesn't depend on X, so
you can install it on machines that don't have X.

You *CANNOT* build Tethereal without GLib; it makes significant use of
GLib facilities, and will continue to do so for the foreseeable future. 
Install it on your headless machine - if you're installing it from a
binary package rather than from source, make sure you install the
development package for it as well (library packages, as prepared for a
number of OSes, include both "user" and "development" packages, where
the latter package installs header files and non-shared libraries; the
name of the development package probably depends on the OS - note that
Linux, in this case, is not an OS, but a family of OSes, as packages in
different distributions may have different names - and on the person or
organization who produced the package, so I can't help you find the
package in question).
